Effect of T-activin on Cellular Immune Response in Balb/c Mice with L-5178-y Lymphoma

چکیده
T-activin is a complex of immunologically active peptides (MW 1.2-6 KDa) derived from the thymuses of different animal species. To study immunomodulatory and antitumor effect of T-activin and its relation with type 1 and type 2 cytokines, immunosuppressed mice with L-5178-Y lymphoma were treated with subcutaneous T-activin. T-activin treatment increased life span and reduced the increase in body weight observed in lymphoma-bearing mice. Effect of T-activin on cellular immune response was evaluated with delayed hypersensitivity test to DNFB and splenic lymphoproliferative response. Both depressed immune response to DNFB and depressed proliferative response observed in lymphoma-bearing mice were totally reverted with T-activin treatment to values observed in control mice treated. In order to know the relation between the above results and cytokine production and T cell subsets, concentrations of IFN- and IL-4 in culture supernatants were determined by ELISA and T cell subsets were determined using flow cytometric analysis. Recovery was not observed in IL-4 levels but complete recovery was observed in IFN- levels. On the other hand, T-activin reversed the lymphoma-induced increment of CD4+ T cells and with this the CD4/CD8 ratio was restored to control value. These findings suggest that the mechanism through which T-activin exerts its antitumor effect is by inducing a type 1 response through IFN- production, thereby promoting cellular immune response.

The role of surface charge of ISCOMATRIX nanoparticles on the type of immune response generated against Leishmaniasis in BALB/c mice
Objective(s): ISCOMATRIX vaccines have now been shown to induce strong antigen-specific cellular or humoral immune responses to a broad range of antigens of viral, bacterial, parasite or tumor. In the present study, we investigated the role of ISCOMATRIX charge in induction of a Th1 type of immune response and protection against Leishmania major infection in BALB/c mice. Materials and Methods:...
متن کاملInduction of Immunogenic Response in BALB/c Mice by Live and Killed Form of Recombinant Lactococcus lactis Displaying EG95 of Echinococcus granulosus Immunogenic Response in BALB/c Mice
Background: CE is a zoonotic parasitic infection caused by Echinococcus granulosus worldwide and is associated with economic losses among livestock animals. EG95 is an immunogenic antigen from the E. granulosus. Lactococcus lactis has been prested as a safe vehicle for antigen delivery. The goal of this study was to design a novel L. lactis strain displaying EG95 as a vaccine delivery system. M...
متن کاملWestern Blot Analysis of Sera from Leishmania major-Infected BALB/c and C57BL/6 Mice
In Leishmania (L.) major-infected BALB/c mice Th2-type cells results in disease progression, whereas C57BL/6-infected mice mount a Th1-type response, which leads to control of the infection. Th2 response correlates with IgG1 whereas, Th1 response supports switching to IgG2a subclasses. Since IgG isotype-dominated response depends on different CD4 + T cell subsets, we studied the antigenic profi...

Introduction: Saffron (thread-like reddish colored stigmas of Crocus sativus L. flowers) was used in traditional medicine for cancers and infectious diseases. Recently, anti-tumor, cytotoxic, anti-mutagenesis and antioxidant activities of saffron metabolites, especially its carotenoids (crocins and crocetin), have been reported. Objective: Since the saffron is used as a spice for food flavo...
متن کاملEffect of IL-2 co-expressed or co-inoculated with immuno-dominant epitopes from VP1 protein of FMD virus on immune responses in BALB/c mice
Objective(s): The results of studies on vaccine development for foot-and-mouth disease (FMD) virus show that the use of inactivated vaccines for FMD virus is not completely effective. Novel vaccinations based on immuno-dominant epitopes have been shown to induce immune responses. Furthermore, for safety of immunization, access to efficient adjuvants against FMD virus seems to be critical.Materi...
